Research summary
Our research interests lie in understanding how variations in DNA structure can affect fundamental biological processes such as replication and transcription. We are motivated by determining biomolecular mechanisms of action, with a long-term view to improving the development of therapeutics. Our research uses high-resolution atomic force microscopy methods Alice pioneered in collaboration with Bruker (CA, USA). To quantify and analyse our high resolution data, we are developing TopoStats, a Python pipeline to automate processing of AFM data from any molecular system. We are developing tools for TopoStats which automate identification and structural characterisation of complex DNA molecules as they interact with proteins and small molecules.
Our research is carried out with our network of collaborators who are experts in atomic force microscopy development, complementary single molecule biophysics techniques, machine learning, molecular dynamics simulations and biochemistry. Together we work to uncover the hidden effect of topological stress in the genome on interactions across its entire conformational landscape. We hope that this knowledge can be harnessed to combat disease by aiding in the rational design of novel therapeutics.
